Effect of Quinidine on the Potassium Permeability and Calcium Binding of the Rabbit Erythrocyte Membrane

Abstract
|
|
|
This study was undertaken to investigate the effect of quinidine on the K permeability and Ca binding of the rabbit erythrocyte membrane. The results are summarized as follows.
1. Quinidine causes the efflux of K across the rabbit erythrocyte membrane. The osmotic resistance of quinidine-treated erythrocytes is diminished. This diminution of the osmotic resistance is more pronounced by increasing concentration of quinidine and longer incubation time. But osmotic resist¡©ance is rather increased in the presence of lower concentration of quinidine.
2. Cysteine and glutathione have little influence on the diminution of osmotic resistance and K efflux induced by quinidine.
3. The addition of PCMB and DNFB augments K outflux markedly while EDTA and EGTA fail to protect against the K efflux induced by quinidine.
4. Quinidine inhibits Na-K ATPase activity of ghosts cells of the rabbit. The inhibition of Na-K ATPase activity by quinidine is not prevented by an addition of cysteine in the incubation medium. 5. Quinidine inhibits Ca binding competitively to rabbit erythrocyte membrane fragments.
